Do I have to do a JSA in order to develop a safety manual in my workplace??

I am a trainee in a company, the boss asekd me to buid a safety manual.

Have fun. You have to KNOW the OSHA standards to develop a safety manual. BTW if you make a mistake that results in injury or death in the manual (that is not in OSHA guidelines), your personally responsible, and may have legal issues.

A JSA is not required. Also, if you really don't know the task that well, the people who do the task are the best ones to do a JSA on that task. But JSAs and JHAs are not required unless your facility wants to be in VPP. There is a publication by one of the state-run OSHA programs that summarized every rule that has a written program requirement.
