You've given a very good idea of what you do, but this will be in your resume or CV.

Dear Madam
In reply to your advertisement in the Washington Bugle of 10 October, please consider my application for the vacancy of Bookkeeper - Fulltime.
With broad experience in Accounts Payable...(etc)... and current part time employment at XYZ, I am applying for your position as it would suit my need for local full-time employment. I would bring to this position strong skills in Excel, MYOB, Quicken, and salary and conditions administration.
Please find attached my brief resume, which I look forward to expanding at interview - for which I am available any day, but would prefer Tuesdays as I am not at XYZ that day. My current employer is aware that I am writing to you, and is happy to give me a firm reference.
I look forward to meeting you to discuss my application
Yours faithfully

used to read hundreds of applications. The ones that gave a good reason to be considered, in the cover letter, usually got an interview!

You don't need the ' at the end of Bookkeeper or you need to change it to Bookkeeper's placement.
Also the " after position needs to go...
Sale Ledger Clerk should be Sales Ledger.
Other than that it looks good!! Best of luck with the job - I'm currently studying an accounts qualification and work in Finance and Payroll - it takes a special breed doesn't it??!!
xx

It's OK, but could be improved. You have some incomplete sentences. Also, the purpose of a cover letter to highlight points in your resume. It should be a sales letter for your resume and you. And it should address specific point in the ad you're responding to. See my quick rewrite below.

Specifically, you asked for someone with [list at least one job requirement specified in the ad--maybe "x years of experience.] I have [now specifically address that requirement--such as "I have x years of experience as a..."]

The main thing to try to do is to show them that you're a perfect match for what they're looking for. So, to the extent you can, cite requirements from their ad and show how you meet them.
Include your contact information in the letter because sometimes letters and resumes get separated.
Hope that helps.
